The research group Struc­tural Bio­chem­istry (Prof. Markus C. Wahl, PhD; https://www.bcp.fu-berlin.de/en/chemie/biochemie/research-groups/wahl-group ) elu­cid­ates the 3D atomic struc­tures of bio­lo­gical mac­ro­molec­ules / bio­molecu­lar com­plexes involved in proka­ryotic and euk­a­ryotic gene expres­sion pro­cesses, and invest­ig­ates the bio­lo­gical activ­it­ies of these molecules / com­plexes via struc­ture-informed func­tional ana­lyses. The exper­i­mental approaches include recom­bin­ant pro­duc­tion of com­pon­ents of the invest­ig­ated molecu­lar machiner­ies, recon­sti­t­u­tion of bio­molecu­lar com­plexes, puri­fic­a­tion of bio­molecu­lar com­plexes from nat­ive sources, bio­chem­ical and bio­phys­ical char­ac­ter­iz­a­tion of bio­molecu­lar com­plexes and their com­pon­ents, molecu­lar struc­ture ana­lysis via integ­rat­ive struc­tural bio­chem­ical approaches (includ­ing mac­ro­molec­u­lar crys­tal­lo­graphy and single-particle cryo­genic elec­tron micro­scopy) as well as the devel­op­ment and applic­a­tion of struc­ture-informed, func­tional assays in vitro and in vivo.

Work­ing field:

Par­ti­cip­a­tion in pro­ject C08 “Tran­scrip­tional and trans­la­tional mech­an­isms of star­va­tion prim­ing in Sta­phyl­o­coc­cus aureus” of Col­lab­or­at­ive Research Cen­ter (CRC) 973 (“Prim­ing and Memory of Organ­is­mic Responses to Stress”; http://www.sfb973.de/ ), with the oppor­tun­ity to obtain a doc­toral degree.

Pro­ject C08 invest­ig­ates non-spe­cific tol­er­ance to redox-act­ive com­pounds and anti­bi­ot­ics dur­ing entry into sta­tion­ary phase (“star­va­tion prim­ing”) in the import­ant human patho­gen Sta­phyl­o­coc­cus aureus. The suc­cess­ful applic­ant will struc­tur­ally ana­lyze tran­scrip­tion com­plexes rel­ev­ant to star­va­tion prim­ing, and assess the impact of these com­plexes on star­va­tion prim­ing by struc­ture-informed func­tional assays in vitro and in vivo. The work will be con­duc­ted in close col­lab­or­a­tion with the research group Molecu­lar Micro­bi­o­logy (Prof. Dr. Haike Antel­mann; https://www.bcp.fu-berlin.de/en/biologie/arbeitsgruppen/mikrobiologie/ag_antelmann ).

Require­ments:

Uni­versity degree (Dip­lom / Mas­ter of Sci­ence) in Bio­chem­istry or a related dis­cip­line with excel­lent grades

Desir­able:

The­or­et­ical know­ledge in and, ideally, hands-on exper­i­ence with
  • micro­bi­o­lo­gical tech­niques
  • molecu­lar bio­logy tech­niques, molecu­lar clon­ing
  • recom­bin­ant pro­duc­tion and puri­fic­a­tion of pro­teins / pro­tein com­plexes using nat­ive pro­ced­ures
  • char­ac­ter­iz­a­tion of pro­teins / pro­tein com­plexes using bio­chem­ical and bio­phys­ical meth­ods (e.g. CD spec­tro­scopy, ther­mo­fluor ana­lysis, gel fil­tra­tion chro­ma­to­graphy / multi-angle light scat­ter­ing, sur­face plas­mon res­on­ance, iso­thermal titra­tion calor­i­metry, multi-scale ther­mo­phoresis)
  • ana­lysis of enzymes via activ­ity assays

  • struc­tural bio­logy meth­ods (e.g. mac­ro­molec­u­lar crys­tal­lo­graphy, NMR spec­tro­scopy, cryo­genic elec­tron micro­scopy and com­ple­ment­ary meth­ods)

  • bac­terial gene expres­sion pro­cesses



This pos­i­tion is embed­ded in the gradu­ate school of the CRC 973 and offers a wide range of oppor­tun­it­ies for col­lab­or­a­tion, train­ing and per­sonal devel­op­ment.
